In indirect immunohistochemistry, the first (primary antibody) binds to the desired antigen and the second one (secondary antibody) has the enzyme attached and it binds to the primary antibody. A substrate is then introduced that the enzyme converts to a colored product.
Often this is combined with a counter-stain for context (e.g. hematoxylin to stain cell nuclei blue).
